No. Label Sync is additive-only by default — it creates missing labels on target boards but never removes any. If you want to clean up orphaned labels, you can explicitly enable deletion in the settings, and you'll always see a preview of what would be removed before anything happens.

Each board you open the Power-Up on acts as a source for its own sync group. So you can set up multiple sync groups by configuring Label Sync on different source boards — each with its own set of target boards.
